y - Glutamyl transferase isoenzymes in human bile

چکیده

The y-glutamyl transferase isoenzymes of bile were studied using electrophoretic, gel filtration, and ultracentrifugation techniques. In view of the known association of other biliary enzymes with lipids the effects of butanol extraction were investigated. The results show the presence of four isoenzymes ofy-glutamyl transferase in bile, differing in electrophoretic mobilities, molecular size, and density. The correlation between the properties of biliary y-glutamyl transferase and of alkaline phosphatase is discussed.

Clinical assessment of the electrophoretic separation of alkaline phosphatase isoenzymes.

Over a five-month period, using data from patients in whom alkaline phosphatase (ALP) isoenzyme studies were requested routinely, we compared actual clinical diagnoses with the predicted diagnoses based on the results of electrophoretic separation of ALP isoenzymes on cellulose acetate before and after heat treatment and on elevated enzymatic activity of gamma glutamyl transferase (GGT) and ala...

Inhibition of glutathione S-transferase by bile acids.

The effects of bile acids on the detoxification of compounds by glutathione conjugation have been investigated. Bile acids were found to inhibit the total soluble-fraction glutathione S-transferase activity from rat liver, as assayed with four different acceptor substrates. Dihydroxy bile acids were more inhibitory than trihydroxy bile acids, and conjugated bile acids were generally less inhibi...

Hepatocarcinogenesis Initiated by Diethylnitrosamine Promotive Effects of Steroids and Bile Acids on

The ability of four steroids and three bile acids to promote the appearance of hyperplastic foci and hyperplastic nodules initiated by diethylnitrosamine (DEN) in the liver of male Fischer rats was tested and compared with that of the known promoter phénobarbital (PB, 0.05%) as a standard value. Two weeks after a single dose of 200 mg of DEN per kg, the animals were exposed to test chemicals f...

Mutations in the nuclear bile acid receptor FXR cause progressive familial intrahepatic cholestasis

Neonatal cholestasis is a potentially life-threatening condition requiring prompt diagnosis. Mutations in several different genes can cause progressive familial intrahepatic cholestasis, but known genes cannot account for all familial cases. Here we report four individuals from two unrelated families with neonatal cholestasis and mutations in NR1H4, which encodes the farnesoid X receptor (FXR),...

-Glutamyltransferase: value of its measurement in paediatrics

-Glutamyltransferase (GGT) is a microsomal enzyme that is widely distributed in human tissues involved in secretory and absorptive processes, particularly the bile canaliculi. Serum GGT is elevated in liver diseases affecting the biliary system, such as extrahepatic biliary atresia, sclerosing cholangitis and progressive familial intrahepatic cholestasis (PFIC) type 3. Conversely, two other sub...
